Comfort, of course, varies from person to person, but good things to look out for are whether or not the shoe you're purchasing comes in varying widths, whether it has a high heel drop and arch support, and what kind of footbed it has. The more of any of these, the merrier (and comfier) the shoe is likely to be.
Keeping in mind where and when you're most likely to wear these shoes will also help you out in the long run. Loafers, for example, are far more versatile and easy to dress up or down than, say, brogues. You can get away with wearing one of them on a casual day much more than the other.
